SYBR GREEN qPCR · NO ROX

Add primers and template. Everything else is in the mix.

Biomiga's SB-Green 2X qPCR Master Mix (QP1311) is a ready-to-use SYBR Green I master mix for real-time quantitative PCR. It contains hot-start Taq DNA polymerase, dNTPs, Mg²⁺, SYBR Green I dye, enhancer and stabilizers. It contains no internal reference dye, for real-time instruments that do not require ROX. The hot-start Taq is antibody-inactivated, which blocks polymerase activity at room temperature; activity is restored once the reaction reaches 95 °C.

How it works

SYBR Green qPCR workflow with standard and fast cycling conditions for Biomiga 2X qPCR master mixes

Biomiga's suggested starting protocol. Optimal conditions vary with primer set.

STEP 1

Thaw and mix

Thaw at room temperature, mix gently, spin down and keep on ice.

STEP 2

Assemble

Combine the 2X mix, primers, template and water on ice or at room temperature, then seal with optical caps or film and spin down.

STEP 3

Activate and cycle

95 °C for 2 min, then 30–45 cycles of 95 °C and 60 °C, collecting data at 60 °C.

STEP 4

Melt curve

Biomiga recommends a melt curve on every run to check for primer dimers and confirm specificity.

Reactions scale from 5 to 50 µL; keep all volumes proportional. Adding template in 5–10 µL improves precision, and undiluted cDNA should be no more than 10% of the reaction.

Primers

Design for 70–150 bp amplicons. 200 nM per primer suits most reactions; titrate between 100 and 500 nM if needed.

Magnesium

3 mM final works for most targets; the optimum can be 3–6 mM. Add 1, 2 or 3 µL of 50 mM MgCl₂ per 50 µL to reach 4, 5 or 6 mM (MgCl₂ not included).

Match the mix to your instrument's ROX requirement. Can I use this mix with TaqMan or other probes?

No. These mixes use SYBR Green I dye-based detection. For probe assays, ask us about Biomiga's Probe-based 2X qPCR Master Mix (QP1314).

What do I need to add?

Primers, template and nuclease-free water. Add 50 mM MgCl₂ only if you want more than the built-in 3 mM final.

Does it support fast cycling?

Yes. After the 2-minute activation at 95 °C, Biomiga's fast protocol uses 95 °C for 3–5 s and 60 °C for 20–30 s per cycle.

How should it be stored?

At −20 °C, protected from light. For convenience it can be kept at 4 °C for up to 6 months. Avoid repeated freeze-thaw, although Biomiga reports no significant performance loss after 10 cycles.

What amplicon size works best?

70–150 bp. Biomiga advises keeping amplicons under 500 bp and running a melt curve on every plate to check for primer dimers.

How is the mix qualified?

Biomiga functionally tests the mix in qPCR and reports linear resolution over six orders of magnitude (R² > 0.995) with efficiency above 90%.

How many reactions are in a pack?

100 reactions (2 × 1.25 mL) or 500 reactions (10 × 1.25 mL) of 2X mix, sized for 50 µL reactions. Reactions can be scaled down to 5 µL, which stretches each pack further.

Can I switch from another vendor's SYBR Green mix?

Usually, as long as the ROX level matches your instrument. Biomiga has not published side-by-side data, so confirm primer efficiency on a standard curve before switching an established assay.

    Payment Terms
    Orders are prepaid by card, or invoiced against an institutional purchase order once credit is approved. Prices are in USD and subject to change without notice. Purchase orders are due within 30 days of invoice unless agreed otherwise in writing.

    Order Confirmation & Cancellations
    Confirmed orders are placed with the manufacturer and are final. Changes or cancellations must be requested in writing before shipment and approved by Blue Tiger Scientific; approved cancellations may incur restocking, materials or third-party fees.

    Shipping & Cold Chain
    Shipping is calculated at checkout. Temperature-sensitive products ship by expedited service, and ship dates may be adjusted to avoid weekend or holiday transit. Orders placed on weekends or holidays are processed the next business day. Delivery dates are estimates and are not guaranteed. A recipient must be available to receive and store temperature-sensitive products on arrival.     Receiving & Claims
    Inspect every shipment on arrival. Shortages, damage or thawed temperature-sensitive product must be reported to Blue Tiger Scientific in writing, with photos, within 2 business days of delivery. Claims received after that window, and shipments that are refused, unclaimed or left unattended, are not eligible for credit or replacement.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option. Blue Tiger Scientific coordinates these claims with Biomiga. No other warranties, express or implied, are given.

    Limitation of Liability
    Blue Tiger Scientific's total liability for any claim is limited to the purchase price of the product involved. Blue Tiger Scientific and Biomiga are not liable for indirect, incidental or consequential damages, including lost research time or materials, arising from the use or misuse of any product.
