TOTAL RNA WITH gDNA REMOVAL

Genomic DNA cleared before the RNA ever reaches the column.

The Biomiga Cell/Tissue Total RNA Extraction Kit Plus (R6321) lyses samples in denaturing Buffer HLY, which Biomiga says immediately inhibits RNase activity. The lysate passes through a DNA Clearance column that traps genomic DNA. The flow-through is mixed with ethanol, bound to an ezBind RNA column, washed twice, and eluted in 50–100 µL of DEPC-treated water. Biomiga states the protocol takes under 30 minutes and describes the RNA as ready for NGS, RT-PCR, qPCR, differential display and microarrays.

How it works

STEP 1

Lyse

Add 500 µL Buffer HLY (with β-mercaptoethanol added) and homogenize. Use 700 µL for more than 5 × 10⁶ cells or more than 15 mg tissue.

STEP 2

Clear gDNA

Spin the lysate through the DNA Clearance column. Keep the flow-through; the genomic DNA stays on the column.

STEP 3

Bind and wash

Add half a volume of 100% ethanol, bind to the ezBind RNA column, then wash with Buffer HRB and RNA Wash Buffer. Finish with a dry spin.

STEP 4

Elute

Elute in 50–100 µL of DEPC-treated water. Biomiga notes that re-applying the eluate typically recovers another 20–30% of the RNA.

Don't exceed 30 mg of tissue or 1 × 10⁷ cells per column; overloading lowers yield and lets genomic DNA through.

What to expect from the RNA

Size range

RNA shorter than about 200 bases doesn't bind efficiently, so this kit isn't suited to miRNA or other small-RNA work.

Genomic DNA

Biomiga states that the clearance column and RNA column together remove nearly all genomic DNA, so DNase digestion isn't needed for most applications. Sensitive applications may need an additional DNase I digest; Biomiga's DNase I (E11-01) is sold separately.

Getting the most RNA

Do

  • Add β-mercaptoethanol (or 1 M DTT) to the Buffer HLY you'll use, as the manual describes; it then keeps up to 1 month at room temperature
  • Warm Buffer HLY to 37 °C if crystals form
  • Add ethanol to the RNA Wash Buffer before first use
  • Remove all culture medium before lysis
  • Stabilize tissue in RNAsecure solution or snap-freeze it

Don't

  • Exceed 30 mg tissue or 1 × 10⁷ cells per column
  • Skip the dry spin before elution
  • Add bleach or acidic solutions to the preparation waste; Biomiga's manual notes Buffer HLY contains chaotropic salts

How is genomic DNA removed?

What samples can I use, and how much?

Cultured cells (suspension or adherent) and animal tissue. Use 500 µL Buffer HLY for up to 5 × 10⁶ cells or 15 mg tissue, and 700 µL for up to 1 × 10⁷ cells or 30 mg tissue. Don't exceed 1 × 10⁷ cells or 30 mg tissue per column; overloading causes genomic DNA carryover.

What do I need to prepare before first use?

Add β-mercaptoethanol (or 1 M DTT) to the volume of Buffer HLY you'll use, as described in the user manual; once added, Buffer HLY keeps at room temperature for up to 1 month. Add 100% ethanol to the RNA Wash Buffer: 8 mL for the 4-prep kit, 48 mL for the 50-prep kit, or 200 mL for the 250-prep kit. If crystals form in Buffer HLY, dissolve them at 37 °C before use.

How long does a prep take?

Biomiga states the protocol can be completed in under 30 minutes. Tissue homogenization time varies by sample.

How should the kit be stored and handled?

All components store at room temperature. Store purified RNA at −20 °C or colder. The expiry date is printed on the label. Biomiga's manual advises gloves and eye protection when handling Buffers HLY and HRB, and not adding bleach or acidic solutions to the preparation waste.

How does it compare with the QIAGEN RNeasy Plus Mini Kit?

Both are silica spin-column kits that remove genomic DNA on a dedicated column before RNA binding. Both retain RNA longer than about 200 nucleotides, and both list a maximum of 1 × 10⁷ cells or 30 mg tissue per prep. QIAGEN publishes up to 100 µg total RNA per RNeasy Plus Mini column and a protocol under 25 minutes. Biomiga states under 30 minutes for R6321 and doesn't publish a column capacity, so we don't compare capacity or yield. Buffers and protocols differ, so validate on your own samples; the 4-prep trial is sized for that. RNeasy and QIAGEN are trademarks of QIAGEN; Blue Tiger Scientific is not affiliated with QIAGEN.

    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery.
