Total RNA from cells and tissue in a single spin-column run.

The Biomiga Cell & Tissue Total RNA Extraction Kit (R6311) isolates total RNA from cultured cells and animal tissue without phenol/chloroform extraction. Samples are lysed in Buffer LY, bound to an RNA column with ethanol, washed, and eluted in 50–100 µL of DEPC-treated water. Biomiga describes the RNA as ready for RT-PCR, real-time PCR, Northern analysis and microarray work, and especially suited to quantitative gene-expression studies.

How it works

STEP 1

Lyse

Add 500 µL Buffer LY (with β-mercaptoethanol added) to the cell pellet or well. Homogenize tissue in Buffer LY with a rotor-stator or ultrasonic homogenizer.

STEP 2

Bind

Add half a volume of 100% ethanol to the lysate, then spin it through the RNA column.

STEP 3

Wash

Wash with Buffer RB, then RNA Wash Buffer. An optional on-column DNase I step fits in between. Finish with a dry spin, lid open.

STEP 4

Elute

Add 50–100 µL of DEPC-treated water and spin. Store the RNA at −20 °C.

Using more tissue than recommended can exceed the column's capacity, which lowers yield and raises genomic DNA carryover.

What to expect from the RNA

Size range

RNA shorter than about 200 bases doesn't bind efficiently, so this kit isn't suited to miRNA or other small-RNA work.

Genomic DNA

Biomiga notes that only trace genomic DNA remains. The optional on-column DNase I step removes it when your assay needs that. DNase I is not included; Biomiga sells it separately.

Getting the most RNA

Do

  • Add 1% β-mercaptoethanol to Buffer LY before use, then keep it at 4 °C
  • Add ethanol to the RNA Wash Buffer before first use
  • Snap-freeze tissue you can't process right away
  • Check RNA integrity on a denaturing gel before critical downstream work

Don't

  • Exceed the input in the table above
  • Skip the lid-open dry spin
  • Load excess RNA into RT-PCR if genomic DNA signal is a problem; Biomiga suggests 50–100 ng

BEFORE YOU START

100% ethanol, β-mercaptoethanol, a microcentrifuge, a tissue homogenizer (for tissue samples), RNase-free DNase I (optional), and pipettes with RNase-free filter tips.

Does it recover miRNA or other small RNAs?

No. Biomiga notes that RNA shorter than about 200 bases doesn't bind the column efficiently, so this kit isn't suited to small-RNA work.

Is the RNA free of genomic DNA?

Not guaranteed. Biomiga notes trace genomic DNA may remain. You can add the optional on-column DNase I step (DNase sold separately), or choose R6321 Plus, which includes a gDNA clearance column.

What sample types does it handle?

Suspension and adherent cultured cells, and animal tissues such as liver, kidney, spleen, lung, heart, muscle, brain and pancreas. Biomiga's yield table also includes tomato leaves. Ask us before using other plant tissues.

What do I need to prepare before first use?

Add 1% β-mercaptoethanol to Buffer LY and store it at 4 °C. Add 100% ethanol to the RNA Wash Buffer: 48 mL for the 50-prep kit, 200 mL for the 250-prep kit.

How long does a prep take?

Biomiga describes the method as taking about 30 minutes. The optional DNase step adds a 15-minute incubation, and tissue homogenization time varies by sample.

How should the kit be stored?

Kit components store at room temperature. Once β-mercaptoethanol is added, keep Buffer LY at 4 °C. Store purified RNA at −20 °C or colder. The expiry date is printed on the label.

How does it compare with the QIAGEN RNeasy Mini Kit?

Both are silica spin-column kits for total RNA from cells and animal tissue, and both are designed around RNA longer than about 200 nucleotides, so neither is a small-RNA kit. Neither removes genomic DNA in its standard protocol without an optional DNase step. QIAGEN publishes a binding capacity of up to 100 µg per RNeasy Mini column; Biomiga's R6311 manual doesn't state a column capacity, so we don't compare capacity or yield. The two kits use different buffers and protocols, so validate R6311 on your own samples first; the 4-prep trial pack is sized for that. RNeasy and QIAGEN are trademarks of QIAGEN; Blue Tiger Scientific is not affiliated with QIAGEN.

Can this kit be used for clinical or diagnostic work?

No. It is intended for in vitro research use only and is not for use in diagnostic procedures or in humans.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option. Blue Tiger Scientific coordinates these claims with Biomiga. No other warranties, express or implied, are given.
