BIOMIGA PCR ENZYMES

A high-fidelity PCR enzyme with everything you need to tune it.

Express Hi Fi DNA Polymerase from Biomiga ships with its own 5X reaction buffer, a separate 20 mM MgCl₂ stock and DMSO, so you can run Biomiga's routine protocol as written or adjust magnesium and additives for a difficult template. The 1X buffer delivers 1.5 mM Mg²⁺; Biomiga recommends optimizing in 0.2 mM steps using the supplied MgCl₂. Extension runs at 72 °C for 10 seconds per kb, and the PCR products have blunt ends.

STEP 1

Assemble on ice

Combine 5X buffer, dNTPs, primers, template and optional DMSO; add the enzyme last and mix gently.

STEP 2

Start hot

Move tubes straight from ice into a thermocycler already at denaturation temperature.

STEP 3

Cycle

95 °C for 30 s, then 25–35 cycles of 95 °C, 52–68 °C and 72 °C at 10 s per kb; finish at 72 °C for 8 min.

Clone blunt

Biomiga notes the products have blunt ends and recommends blunt-end cloning.

Primers and Mg²⁺

Primers can run at 0.2–1 µM final. Mg²⁺ is affected by dNTPs, template, additives and EDTA; adjust in 0.2 mM steps with the supplied MgCl₂.

What comes in each pack?

Every pack contains the enzyme at 2 U/µL, 5X Express Hi Fi Buffer, 20 mM MgCl₂ and DMSO. EHF1101-01 has 500 U (250 µL) of enzyme; EHF1101-02 has 2,500 U (1,250 µL), with buffer, MgCl₂ and DMSO scaled up to match. See the box contents table above.

What kind of ends do the PCR products have?

Biomiga states the products have blunt ends and recommends blunt-end cloning. Blunt products lack the single 3′ A overhang that TA-cloning vectors need.

About how many reactions does a pack run?

At the IFU's 1 µL of enzyme per 50 µL reaction, the enzyme volume works out to roughly 250 reactions (EHF1101-01) or 1,250 (EHF1101-02); the buffer at 10 µL per reaction covers about 240 or 1,200. Smaller 20 µL reactions stretch both further. These are approximate divisions of Biomiga's volumes, not a stated reaction count.

Do I need to add magnesium?

Not for a standard run: the 1X buffer already gives 1.5 mM Mg²⁺. Biomiga notes that dNTP concentration, the template, additives and any EDTA affect the optimum, and suggests adjusting in 0.2 mM steps with the MgCl₂ provided.

When should I use the DMSO?

For GC-rich templates or sequences with strong secondary structure. Follow the current instructions for use for the final concentration.

How much template should I use?

For a 50 µL reaction, Biomiga suggests 50–250 ng of genomic DNA or 10–50 ng of plasmid or viral DNA. High-quality template improves results.

Can I run it in a protocol written for another vendor's high-fidelity enzyme?

Use Biomiga's buffer, magnesium and cycling conditions rather than another product's. Biomiga has not published a side-by-side comparison, so run a small pilot before switching an established assay.

Is there a ready-to-use version?

    Receiving & Claims
    Inspect every shipment on arrival. Shortages, damage or thawed temperature-sensitive product must be reported to Blue Tiger Scientific in writing, with photos, within 2 business days of delivery. Claims received after that window, and shipments that are refused, unclaimed or left unattended, are not eligible for credit or replacement.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option.
