WHY ENDOTOXIN-FREE

Clean plasmid for the cells that notice.

Plasmid made with a standard prep carries bacterial endotoxin (lipopolysaccharide) from the E. coli outer membrane. Robust lines like HEK293 tolerate it, but endotoxin-sensitive cell lines, primary cells and microinjection work call for it to be removed first. The Biomiga EndoFree ezFilter Plasmid Maxiprep Kit (PD1522) strips endotoxin inside the purification itself and delivers plasmid below 0.1 EU per µg, eluted directly in 1.5 to 2 mL with no precipitation step.

How it works

STEP 1

Lyse and clear

Resuspend, lyse and neutralize, then push the lysate through the 60 mL ezFilter syringe. About five minutes instead of a 20-minute high-speed spin.

STEP 2

Strip endotoxin and bind

Endotoxin-removal buffer and ethanol go into the cleared lysate, which then loads onto the maxi column. No phase-partition extraction.

STEP 3

Wash and dry

Wash the column and dry it, on a vacuum manifold or in a centrifuge.

STEP 4

Elute

Elute in 1.5 to 2 mL of EndoFree Elution Buffer and reapply once for a second pass.

Expected yield

Yield depends on the plasmid's origin of replication, its size and the host strain. Biomiga's figures per 200 mL of culture:

Getting the most plasmid

Do

  • Use an endA- host such as DH5α or TOP10
  • Grow 12 to 16 hours in LB to an OD600 of 2.0 to 3.0
  • Resuspend the pellet completely before lysis
  • Warm Buffer B1 to redissolve any precipitate, and keep its cap tight
  • Scale culture and buffers up 2 to 3 times for low-copy plasmids
  • Dry the column fully before eluting

Don't

  • Overload the column: biomass (OD600 x mL) is optimal at 450 to 550
  • Vortex after adding B1 or lyse longer than 5 minutes
  • Hold culture at 4 °C overnight; freeze the pellet at -20 °C instead
  • Forget to add RNase A to Buffer A1
  • Use rich media above OD600 3.0

EZFILTER VS. STANDARD MAXI

Why the ezFilter version?

Clearing the lysate is the slowest, most equipment-hungry step of a maxiprep. The ezFilter syringe turns it into a five-minute bench step, so you don't need a high-speed rotor for clearing or wait out a 20-minute spin. With a vacuum manifold, several preps run side by side.

You supply: 100% ethanol, 50 mL conical tubes, and a centrifuge reaching 5,000 x g or a vacuum manifold with a 200 to 600 mbar pump.

Is this an alternative to the QIAGEN EndoFree Plasmid Maxi Kit?

Yes, for labs that want endotoxin-free plasmid at maxi scale. Both build endotoxin removal into the prep. The Biomiga kit uses a binding column on a vacuum manifold or centrifuge and elutes directly; the QIAGEN kit uses anion-exchange chromatography under gravity flow. QIAGEN states a lower endotoxin level (below 0.04 EU/µg versus below 0.1 EU/µg).

How much plasmid DNA will I get?

Biomiga lists 700 to 1,000 µg from 200 mL of culture for a high-copy pUC plasmid, and less for lower-copy vectors. See the yield table above.

How long does the prep take?

Biomiga positions its EndoFree ezFlow line as a prep of under 60 minutes, not counting the overnight culture. Times vary with the protocol and the number of preps.

Do I need a vacuum manifold?

No. The manual includes a centrifuge-only protocol. A manifold makes it easier to run several preps at once.

Can I use it for low-copy plasmids?

Yes. Scale the culture volume and the buffer volumes up 2 to 3 times, as Biomiga recommends.

Is the DNA suitable for primary cells?

Biomiga designed the endotoxin-free DNA for transfection of endotoxin-sensitive cell lines, primary cultured cells and microinjection.

How should the kit be stored?

At room temperature (22 to 25 °C), except Buffer A1 after RNase A is added and Buffer ER, which go to 4 °C. Shelf life is guaranteed for 12 months from purchase.

Can this kit be used for clinical or in vivo human use?

No. It is intended for in vitro research use only and is not for use in humans.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option.
