ONE-STEP RT-PCR

From RNA to PCR product in one tube and one program.

Biomiga's One-Step RT-PCR Kit (RT0201) reverse-transcribes and amplifies a specific RNA target from total RNA or mRNA in a single reaction. It uses a mixture of MMLV reverse transcriptase and hot-start Taq DNA polymerase in an optimized buffer. Biomiga states it can detect RNA targets up to 4.0 kb from 0.01 pg to 2 µg of total RNA. A separate 5 mM magnesium sulfate stock is included for optimization.

How it works

One-step RT-PCR workflow for Biomiga RT0201: assemble on ice, reverse transcribe at 40–50 °C, amplify for 40 cycles, check the product on a gel

Biomiga's suggested starting program, 50 µL reactions.

STEP 1

Assemble on ice

25 µL 2× reaction mix, 1 µL enzyme mix, forward and reverse primers, and RNA template in a 50 µL reaction.

STEP 2

Reverse transcribe

Program the thermal cycler so cDNA synthesis is followed immediately by PCR: 40–50 °C for 10 min.

STEP 3

Amplify

94 °C for 2 min, then 40 cycles of 94 °C for 15 s, 55–65 °C for 30 s and 68 °C for 1 min per kb.

STEP 4

Check on a gel

Analyze the RT-PCR products by gel electrophoresis.

Magnesium

The 2× reaction mix already contains Mg²⁺. If needed, optimize further with the supplied 5 mM magnesium sulfate. Note this is MgSO₄, not MgCl₂.

Detection

The kit produces an end-point PCR product for gel analysis. It contains no fluorescent detection dye, so it is not a real-time qPCR kit.

What is included?

An enzyme mix (MMLV RT and hot-start Taq), a 2× reaction mix containing dNTPs and Mg²⁺, and 5 mM magnesium sulfate. You supply primers, RNA and water.

Can I use it for real-time qPCR?

No. Biomiga's protocol is for end-point detection by gel, and the kit contains no fluorescent dye. For quantitative work, make cDNA with FS01 and use a SYBR Green qPCR mix, or ask us about Biomiga's one-step qRT-PCR mix.

What RT temperature should I use?

Biomiga suggests 40–50 °C for 10 minutes, followed immediately by PCR.

How long should the extension be?

68 °C for 1 minute per kb of target, for 40 cycles.

Why magnesium sulfate?

The 2× reaction mix already contains magnesium. The 5 mM MgSO₄ is supplied for further optimization if needed; use it rather than MgCl₂.

Should I choose this kit or the 1st Strand cDNA Synthesis Kit?

Choose RT0201 for a quick end-point answer on a specific target. Choose FS01 when you want a cDNA pool for qPCR or several assays.

How should the kit be stored?

At −20 °C. Biomiga states the kit is stable for 2 years when stored this way.

    Product Use & Eligibility
    Products are for in vitro research use only (RUO). They are not intended for human or animal use, diagnostic procedures, or therapy, and may not be resold to consumers. Sales are limited to verified institutional and business customers shipping to a business address; we do not ship to residential addresses. Blue Tiger Scientific may request institutional verification or end-use information before shipping and may cancel any order that cannot be verified.

    Payment Terms
    Orders are prepaid by card, or invoiced against an institutional purchase order once credit is approved. Prices are in USD and subject to change without notice. Purchase orders are due within 30 days of invoice unless agreed otherwise in writing.

    Order Confirmation & Cancellations
    Confirmed orders are placed with the manufacturer and are final. Changes or cancellations must be requested in writing before shipment and approved by Blue Tiger Scientific; approved cancellations may incur restocking, materials or third-party fees.

    Shipping & Cold Chain
    Shipping is calculated at checkout. Temperature-sensitive products ship by expedited service, and ship dates may be adjusted to avoid weekend or holiday transit. Orders placed on weekends or holidays are processed the next business day. Delivery dates are estimates and are not guaranteed. A recipient must be available to receive and store temperature-sensitive products on arrival.     Receiving & Claims
    Inspect every shipment on arrival. Shortages, damage or thawed temperature-sensitive product must be reported to Blue Tiger Scientific in writing, with photos, within 2 business days of delivery. Claims received after that window, and shipments that are refused, unclaimed or left unattended, are not eligible for credit or replacement.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option. Blue Tiger Scientific coordinates these claims with Biomiga. No other warranties, express or implied, are given.

    Limitation of Liability
    Blue Tiger Scientific's total liability for any claim is limited to the purchase price of the product involved.
