TWO-STEP RT-PCR · cDNA SYNTHESIS

From RNA to PCR-ready cDNA in about an hour.

Biomiga's 1st Strand cDNA Synthesis Kit (FS01) converts total or poly(A)+ RNA into first-strand cDNA. Its reverse transcriptase mix combines an engineered MMLV reverse transcriptase with a ribonuclease inhibitor protein, and the kit supplies both oligo(dT)20 and random hexamer annealing buffers. Biomiga states the system works from 10 pg to 5 µg of total RNA and detects RNA targets from 100 bp to 10 kb, and that it is suited to gene isolation, high-throughput expression profiling by real-time quantitation, and end-point RT-PCR.

How it works

First-strand cDNA synthesis workflow for Biomiga FS01: anneal primer at 65 °C, add RT mix, incubate at 37 °C for 50 min, use 1–2 µL in PCR or qPCR

Biomiga's 1st-strand protocol, 20 µL per reaction.

STEP 1

Anneal primer

Combine RNA with 5 µL annealing buffer OT (oligo(dT)20) or RH (random hexamer), water to 10 µL. Heat at 65 °C for 3 min, then place on ice.

STEP 2

Add RT mix

Add 10 µL of reaction mixture (9 µL cDNA synthesis buffer + 1 µL RT and RNase inhibitor mix). Mix gently and spin briefly.

STEP 3

Reverse transcribe

37 °C for 50 min. With random hexamers, incubate 5 min at room temperature (22–24 °C) first.

STEP 4

Use or store

Optionally heat to 85 °C for 5 min. Use 1–2 µL of the 20 µL reaction per PCR; store the rest at −20 °C.

For mRNA targets under 1.5 kb, a 30-minute incubation is an option. RT temperature can be optimized between 37 and 42 °C. Heat inactivation at 85 °C is recommended when using less than 10 ng of RNA.

No-RT control

For accurate expression profiling, run the same amount of RNA without reverse transcriptase. Any signal in that control points to genomic DNA contamination.

DNase option

If the no-RT control shows product, Biomiga describes an optional DNase I treatment before cDNA synthesis. DNase I and its buffer are not included.

For quantitative results, run FS01 cDNA with a Biomiga SYBR Green qPCR mix matched to your instrument: no ROX (QP1311), low ROX (QP1312) or ROX (QP1313).

Should I use oligo(dT) or random hexamers?

Oligo(dT)20 primes from the poly(A) tail of mRNA. Random hexamers prime throughout the RNA, including transcripts without a poly(A) tail. Both annealing buffers are included, so you can choose per experiment.

How much RNA can I use?

From 10 pg to 5 µg of total RNA per 20 µL reaction, per Biomiga. Poly(A)+ purified RNA can also be used.

How long does it take?

About an hour: 3 minutes at 65 °C to anneal, then 50 minutes at 37 °C (plus 5 minutes at room temperature first with random hexamers). For mRNA targets under 1.5 kb, 30 minutes at 37 °C is an option.

How much cDNA goes into a PCR?

Biomiga recommends 1–2 µL of the 20 µL first-strand reaction per PCR. Store the rest at −20 °C.

Do I need to heat-inactivate the RT?

It is optional. Biomiga recommends 85 °C for 5 minutes when the reaction used less than 10 ng of RNA.

How do I check for genomic DNA contamination?

Run a no-RT control with the same amount of RNA but no reverse transcriptase. If it gives a signal, treat the RNA with RNase-free DNase I before cDNA synthesis, or start from an RNA kit with gDNA removal such as R6321.

Is DNase I included?

No. Biomiga describes an optional DNase I step, but DNase I and its reaction buffer are supplied by you.

Should I choose this kit or the One-Step RT-PCR Kit?

Choose FS01 when you want a cDNA pool to use in several assays or in qPCR. Choose RT0201 when you want to go from RNA to an end-point PCR product in one tube.

Can I switch from another vendor's cDNA synthesis kit?

Usually, but Biomiga has not published side-by-side data. Run a small pilot with a no-RT control before switching an established assay.

How should the kit be stored?

At −20 °C in a non-frost-free freezer. Biomiga states the kit is stable for 2 years when stored this way.

    Payment Terms
    Orders are prepaid by card, or invoiced against an institutional purchase order once credit is approved. Prices are in USD and subject to change without notice. Purchase orders are due within 30 days of invoice unless agreed otherwise in writing.

    Shipping & Cold Chain
    Shipping is calculated at checkout. Temperature-sensitive products ship by expedited service, and ship dates may be adjusted to avoid weekend or holiday transit. A recipient must be available to receive and store temperature-sensitive products on arrival.     Receiving & Claims
    Inspect every shipment on arrival. Shortages, damage or thawed temperature-sensitive product must be reported to Blue Tiger Scientific in writing, with photos, within 2 business days of delivery. Claims received after that window, and shipments that are refused, unclaimed or left unattended, are not eligible for credit or replacement.

    Returns & Refunds
    Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery. Refunds or replacements are issued only for shipping errors or verified product defects. Temperature-sensitive products are not returnable once delivered. Customers pay return shipping unless the error was ours, and refunds exclude original shipping charges. Approved refunds are processed within 45 business days.

    Warranty
    Biomiga warrants its products to perform as described in their labeling when used in accordance with their instructions; the remedy for a product that does not is replacement, at Biomiga's option. Blue Tiger Scientific coordinates these claims with Biomiga. No other warranties, express or implied, are given.

    Limitation of Liability
    Blue Tiger Scientific's total liability for any claim is limited to the purchase price of the product involved.
