Immobilized DNase I – Research Use Only

Immobilized DNase I is a high-purity, matrix-bound deoxyribonuclease I supplied as a 1 mL suspension (Catalog #B2016385). The enzyme is covalently immobilized on agarose beads, allowing easy removal after use, repeated application, and reduced contamination of samples with free enzyme. It is an excellent tool for complete removal of genomic DNA from RNA preparations, elimination of plasmid DNA in protein samples, prevention of viscosity in cell lysates, and many other nucleic acid decontamination workflows.

Scientific Overview

DNase I is an endonuclease that nonspecifically cleaves single- and double-stranded DNA, producing 5'-phosphorylated oligonucleotides. When immobilized on a solid support, the enzyme retains its activity while offering major practical advantages: it can be easily removed by centrifugation or filtration, reused in multiple cycles, and prevents carry-over of free nuclease into sensitive downstream applications. Immobilized DNase I is particularly valuable for RNA purification (removal of genomic DNA contamination), protein sample cleanup, and viscosity reduction in cell lysates without introducing soluble enzyme that could interfere with subsequent steps.

This Immobilized DNase I is suitable for:

  • Complete removal of DNA from RNA preparations
  • Elimination of plasmid or genomic DNA from protein samples
  • Reduction of viscosity in bacterial or mammalian cell lysates
  • Reusable DNA decontamination workflows
  • Prevention of DNA contamination in sensitive molecular biology assays

Usage & Handling Guidance

Store at room temperature (RT). Before use, gently resuspend the immobilized enzyme. Add an appropriate amount of the suspension to your sample and incubate with gentle mixing (typically 15–60 min at room temperature or 37 °C). Remove the immobilized enzyme by centrifugation or filtration. The support can often be washed and reused.

  • Recommended applications: DNA removal from RNA or protein samples, lysate viscosity reduction
  • Typical usage: 10–50 µL of suspension per mL of sample (optimize for complete DNA digestion)
  • Stability: Stable at room temperature; avoid extreme pH or high salt during storage
  • Handling: Resuspend gently before pipetting; use in DNase-free tubes
