α-Defensin-5 (HD-5) is a human antimicrobial peptide secreted by Paneth cells in the small intestine. It plays a critical role in mucosal immunity by neutralizing pathogens, modulating microbiota composition, and regulating inflammatory responses. This 32-residue peptide contains three disulfide bridges that stabilize its β-sheet-rich structure, enabling potent activity against bacteria, fungi, and viruses.

Researchers use α-Defensin-5 in studies of inflammatory bowel disease (IBD), Crohn’s pathology, gut barrier integrity, and innate immune signaling. It has been shown to suppress colon cancer growth via PI3K pathway inhibition, neutralize Clostridioides difficile toxins, and influence microbial sensing in Vibrio cholerae. Its expression is tightly regulated by microbial metabolites and bitter taste receptors, making it a valuable biomarker and therapeutic candidate in gastrointestinal and metabolic research.
