    Biomiga's Lentivirus Concentration Reagent concentrates recombinant lentivirus from transfected-cell culture supernatant without ultracentrifugation. Mix one volume of Buffer LP with three volumes of clarified supernatant, chill, pellet at 3,000 rpm, and resuspend in Buffer LS. Biomiga reports 50–100-fold concentration with about 60–70% recovery.

LENTIVIRUS CONCENTRATION

Concentrate lentivirus on a benchtop centrifuge, not an ultracentrifuge.

Ultracentrifugation has been the traditional way to separate lentivirus from cellular proteins and media, but Biomiga notes it is time-consuming and limits how much material you can process. The Biomiga Lentivirus Concentration Reagent (V2001) concentrates recombinant lentivirus from transfected-cell culture supernatant with two buffers and low-speed spins. Biomiga reports the virus can be concentrated 50–100-fold, with a recovery rate of around 60–70%. Biomiga states the virus is stable in Buffer LP, so the incubation can run overnight.

How it works

STEP 1

Clarify

Spin the culture media 10 min at 3,000 rpm, 4 °C, and pass the supernatant through a 0.45 µm filter. Clarified supernatant can be stored at −80 °C until you're ready.

STEP 2

Add Buffer LP

Mix 1 volume of Buffer LP with 3 volumes of supernatant (for example, 5 mL into 15 mL). Incubate at 4 °C for at least 4 hours or overnight.

STEP 3

Pellet

Spin 30 min at 3,000 rpm, 4 °C. Remove all the supernatant; the pellet may look hazy. Keep it on ice.

STEP 4

Resuspend and store

Dissolve the pellet in 300–500 µL Buffer LS, spin 2 min at 8,000 rpm, and transfer the supernatant to a clean vial. Aliquot and store at −80 °C.

Clarified virus supernatant, 0.45 µm filters, sterile conical and microcentrifuge tubes, a refrigerated centrifuge (3,000 rpm), a microcentrifuge (8,000 rpm, 4 °C), cryovials and a BSL-2 workspace.

How much does it concentrate the virus?

Biomiga reports 50–100-fold concentration with a recovery rate of around 60–70%. Results depend on your construct, packaging system and harvest.

Do I need an ultracentrifuge?

No. Biomiga's protocol uses a refrigerated centrifuge at 3,000 rpm and a microcentrifuge at 8,000 rpm, both at 4 °C. Biomiga gives speeds in rpm, so check your rotor's g-force.

How much supernatant can I process?

Up to 30 mL per prep, about 1–2 T75 flasks. Each prep takes one third of its volume in Buffer LP, so 30 mL of supernatant needs 10 mL. Across a whole pack, the supernatant you can treat is three times the Buffer LP volume.

How long does it take?

Plan for an incubation of at least 4 hours, or overnight, at 4 °C, plus about 45 minutes of spins. Clarified supernatant can be stored at −80 °C before you start.

How should the reagent and the virus be stored?

Store both buffers at room temperature (15–25 °C). Aliquot the concentrated virus and store it at −80 °C. The expiry date is on the label.

What biosafety level is required?

Biomiga warns that lentivirus-containing media and concentrated virus can be infectious to humans and animals. All steps must be performed under at least Biosafety Level 2 conditions.

How does it compare with Takara's Lenti-X Concentrator?

Both use 1 volume of reagent per 3 volumes of clarified, 0.45 µm-filtered supernatant, followed by a low-speed spin at 4 °C instead of ultracentrifugation. Takara's published protocol calls for 30 minutes to overnight at 4 °C and a 1,500 × g spin for 45 minutes. Biomiga specifies at least 4 hours to overnight and 30 minutes at 3,000 rpm. We don't compare recovery between the two products; validate on your own supernatant, and the 10 mL pack is sized for a trial.     Returns require a Return Merchandise Authorization (RMA) issued by Blue Tiger Scientific and must be requested within 30 days of delivery.
