Scientific Overview

ACO2 encodes mitochondrial aconitase (aconitate hydratase), the second enzyme of the tricarboxylic acid (TCA) cycle, which catalyzes the stereospecific isomerization of citrate to isocitrate. The enzyme contains an iron–sulfur cluster that is sensitive to oxidative damage, so ACO2 abundance and activity are frequently examined in studies of mitochondrial function and oxidative stress.

ACO2 is located on chromosome 22q13.2. Biallelic pathogenic variants cause infantile cerebellar-retinal degeneration (ICRD) and have also been linked to optic atrophy, making ACO2 a target of interest in mitochondrial disease and neurodegeneration research.
