Scientific Overview

Keyhole limpet hemocyanin (KLH) is an oxygen-carrying metalloprotein from the keyhole limpet (Megathura crenulata) that has been widely used as a carrier protein for immunization. Because of its large size, multiple epitopes, and inherent immunogenicity, KLH conjugated to haptens or peptides is effective for raising high-titer, specific antibodies. Many research antibodies and commercial antibody kits have been developed using KLH conjugates as immunogens.

When researchers generate antibodies against a KLH-conjugated antigen, some of the resulting antibodies recognize the target antigen, but others recognize KLH itself. KLH-conjugated agarose beads provide a simple method to remove or capture anti-KLH antibodies, either to negative-select for antibodies specific to the target hapten or peptide, or to enrich anti-KLH antibodies for analysis.

Key applications include:

  • Depletion of anti-KLH antibodies from hybridoma or polyclonal antibody preparations
  • Purification of antibodies specific to KLH-conjugated antigens
  • Screening of antibody libraries for KLH-binding species
  • Immunological research and antibody characterization

Usage & Handling Guidance

Store at 2–8°C. Before use, gently mix the suspension by inversion (do not vortex) and resuspend the beads. Protect from freeze–thaw cycles.

  • Batch binding: Add the beads to your antibody solution, incubate for 30 minutes to 1 hour at 4°C or room temperature, then separate by settling or brief centrifugation (1,000 × g for 1 minute).
  • Column packing: Alternatively, pack the beads into a column for gravity-flow or low-pressure chromatography applications.
  • Buffer compatibility: Use antibody storage buffers (PBS, TBS, or similar); avoid extreme pH values that may destabilize KLH or agarose.

What You Get

  • 2 mL of keyhole limpet hemocyanin conjugated to agarose beads, supplied as suspension
  • A ready-to-use affinity matrix for antibody screening and purification
  • High binding capacity for anti-KLH antibodies
  • For research use only (RUO)

Why Researchers Choose It

  • KLH is a gold-standard immunogenic carrier widely used in antibody development
  • Agarose support is chemically inert, biocompatible, and suitable for repeated use
  • Convenient suspension format ready for immediate use in batch or column workflows
  • Effective at depleting unwanted anti-KLH reactivity from antibody pools

Frequently Asked Questions (FAQ)

  • What is KLH and why is it used as a carrier protein?
Keyhole limpet hemocyanin is a large (~400 kDa), highly immunogenic oxygen-transport protein. Its size and multiple epitopes make it an excellent carrier for small antigens (haptens and peptides) to enhance immune recognition.
  • How do I use KLH beads to deplete anti-KLH antibodies?
Incubate your antibody solution with the beads for 30 minutes to 1 hour at 4°C or room temperature, then separate the beads by settling or centrifugation. The flow-through or supernatant contains antibodies depleted of anti-KLH reactivity.
  • Can I reuse these beads?
Yes, with care. Gently wash the beads with PBS or your working buffer to remove unbound antibodies, and re-equilibrate before the next use. Repeated freeze–thaw cycles should be avoided.
  • What if I need to capture and enrich anti-KLH antibodies instead of depleting them?
After incubation, elute the antibodies from the beads using a low-pH buffer (e.g., 0.1 M glycine, pH 2.5) or high-salt buffer, depending on the strength of binding you observe.
  • Are there any monoclonal or recombinant alternatives to KLH-based products?
Yes. For some applications, protein G, protein A, or other affinity matrices may offer improved specificity. Contact us to discuss options for your specific experiment.
This product is for Research Use Only (RUO). It is not intended for diagnostic or therapeutic use in humans or animals.
