α-Defensin-6 (HD-6) is a human antimicrobial peptide secreted by Paneth cells in the small intestine. Unlike other defensins, HD-6 exhibits minimal direct bactericidal activity but forms self-assembling nanonets that trap pathogens, preventing epithelial invasion. This unique mechanism makes HD-6 a key player in mucosal barrier defense and innate immunity.

Researchers use α-Defensin-6 to study gut microbiota modulation, intestinal barrier integrity, and host-pathogen interactions. It has been implicated in Crohn’s disease, amyloid inhibition, and antimicrobial protein regulation via FABP4 signaling.
