Scientific Overview

Concanavalin A is one of the most thoroughly characterized lectins. It recognizes terminal and internal α-D-mannosyl and α-D-glucosyl residues and binds the trimannoside core common to N-linked glycans with high affinity; crystal structures of Con A–trimannoside complexes show how all three sugar residues are held in an extended binding cleft. Carbohydrate binding depends on bound manganese and calcium ions.

Texas Red (sulforhodamine 101 derivative) was introduced as a hydrophilic, red-emitting fluorophore designed to be used alongside fluorescein in dual-parameter flow cytometry and fluorescence microscopy. Its emission near 615 nm is well separated from green fluorophores, so Con A – Texas Red can be combined with FITC- or GFP-labeled probes.
