  • pH adjustment: TRIS buffers can be adjusted by changing the ratio of TRIS-HCl (acidic form) to TRIS base (free base) or by adding HCl or NaOH. Typical working range is pH 7–9.
  • Metal ion compatibility: Unlike EDTA or phosphate buffers, TRIS does not chelate Mg²⁺, Ca²⁺, or Zn²⁺, preserving their availability in enzyme reactions.
  • Sterile use: If required, filter the beads in this buffer through 0.22 µm or sterilize by autoclaving at 121°C, 15 psi for 15 minutes.
